There are HTML based formats, IMS CP and SCORM, specifically designed
for transferring course content. SCORM includes interactive quizzes,
which can be completed off-line. The student downloads a SCORM course
module and then send back a file with the quiz results. However, the
implementations I have seen require the student be on-line (briefly) to
download the package and later again to upload the results of the quiz.

I don't know of any systems which work with sneaker-net. Apart from
students in remote areas, this would be useful for prisoners and
military personnel on deployment. These groups have been catered for in
the past with paper based distance education courses. When I signed up
for a DE Masters of Education, one questions on the enrollment form
asked was if I was a prisoner.
The notes for my e-leaning course "ICT Sustainability" are normally
delivered on-line, a week at a time. But they are also available as:

As I am using Moodle Learning Management System, the student can receive
forum posts by email. But they need Internet access to post responses
and submit assignments. It would be technically possible to modify
Moodle so this could be done by file transfer, but as far as I know, no
one has.
